The origins of the villa date back to 1533 when Duke Alessandro de' Medici decided to build a fort to defend the Era Valley and the Valley of Cascina. The castle had four towers at the four corners of the building, had a parade ground, a stable and barracks. It was probably Cosimo I who continued the work and after several changes of ownership between the Gondi family and the family Botti, in 1620 the property went back again in the hands of the Medici. It was the Medici family who in 1637 sign the documents to transfer the ownership of the house and the estate to Senator Filippo Niccolini. The senator became Marquess of Ponsacco and Camugliano. Even today, the Niccolini family is the owner of the villa.

The coat of arms of the family Niccolini is colored blue with a silver-colored leopard and a red slanting stripe. The towers are still present and a long tree-lined avenue creates a view from the entrance gate to the villa. In the main square, once a parade ground, there is a statue of Hercules slaying the Hydra. The sculptor was Giovanni Bandini and the work is the undisputed star of the square in front of the villa. The main entrance is characterized by a double ramp of steps and the main entrance has three arches. Some architects argue that the construction and the structure of Villa Niccolini Camugliano has served as an example for the Villa L'Ambrogiana, which is also characterized by four towers.

The rooms inside are quite stark, except the hall with frescoes of landscapes visible through a mock-Gothic portico. These frescoes date back to the nineteenth century. Particularly interesting also the arches of the eighteenth century barn. The garden was modified by the Niccolini family. The elliptical-shaped square has hedges and busts in marble around it. The villa is still privately owned.
